To add a little more:
Do you like mountains or the sea?
Greenville and Asheville are awesome if you're looking for higher elevation excitement, and Asheville has some interesting, if angry, people.
Charleston is a bit commercial but it has some amazing expanses of natural space and little hole-in-the-wall geek shops, bars, etc, that make up for it. Just be prepared to pay out the nose for parking downtown and that, if you get lost, it's encircled by a road that rounds the peninsula.
Again, avoid Columbia. There is nothing to do, it is basically a working capitol full of boring people and uninspiring landscapes. Even the lakes are artificial and uninspiring. .
