If you plan to play offensive line, you need to train for what they do at that position.
An offensive lineman rarely moves more than ten yards on a single play. Short, powerful sprints with fast response off the snap. Get used to exploding off the line according to the snap count. I used to go out and do sprints after lifting when it was offseason. I'd warmup then run two 100 yard sprints at 70%, about 6 20 yard sprints full out, and 10 10 yard sprints. Sometimes I'd precede these with plyometric stuff, but I never really got into that, so I don't know if it helped much or not.
When making contact for a block, the offensive lineman will usually shoot his hands into the defender's chest and try his best to lock up with him and steer him with his hips and leg drive. You are going to need powerful strength in both upper and lower body to successfully dominate someone at the line of scrimmage.
To do this, hit the weights. Look up "starting strength wiki" and follow their beginner's program. Drink a fuckload of milk and eat everything in sight. If you're playing on the line, you can't let someone else move you, so fat is as much a tool as muscle. That said, don't become a complete lardass. Fat guys that take plays off for "a breather" are worthless cocksuckers. Make sure you can jog thirty minutes straight about three days a week. And by jog, I don't mean fast walking while pretending you're running. make sure you're conditioned enough to stay in the game.
Finally, work on your agility. For this, you're going to need to run some tires, do ladder drills, do the dot drill. Anything that involves running around with precise footplacement and constant changing of direction. Offensive linemen often need to pull in order to make blocks for screens and trap plays. Proper footwork is essential for these plays.
