So I want to get my rats used to being picked up by me.

I mean actual picking them up, not just letting them crawl onto my hand. Right now most of the time when I do it they don't really like it. So how do I go about it? Do I just pick them up like that until they get used to it? How often should I do it?

If they trust you already (don't bite, don't run away, and really do climb onto your hands), then scooping them up with both hands under their feet (not grabbing them about the middle and NEVER by the tail) regularly shouldn't be a problem. Pick them up and immediately hand them a treat while you hold them, and rub them behind the ears while they eat it. Soon they'll love being picked up and held.
